Protein

characteristic-
  • Polymer consists of monomer amino acids
  • C,H,O,N,P,S
  • polar ( contains carboxyl group,COOH and Amino group,NH2 at terminal end)
  • can be globular and fibrous
  • globular: enzymes & hormones
  • fibrous: structural proteins(i.e. collagen &keratin)
  • 1 or more polypeptide
AMINO ACIDS

  1. condensation-> polypeptide (held by peptide bond)
  2. 20 types a.a (different R group)
  3. different properties ( R group)
  4. carbon atoms attached to
  • amino group
  • carboxyl group
  • R group
  • H atom
5. function determined by specific sequences of a.a. in Polypeptide.
6.zwitterions in water
amphoteric ( can be acidic or basic)


Protein
  1. primary structure- polypeptide chain formed by unique sequence of amino acid, (peptide bond)
  2. secondary structure- a-helix &B- pleated P.P chain, - Hydrogen bonding - fold/coil of PP. (repeating configuration)
  3. Tertiary structure- 3D shape of PP, interaction between amino acids & R group i) ionic bond ii) disulphide bridges iii) Hydrogen bond iv) Hydrophobic & Van der waals interactions
  4. Quartenary- aggregation of 2 or more polypeptide , (collagen/haemoglobin)

#Protein conformation is determined by chemical& physical conditions in protein environment

# Denaturation= Protein unravels & lose native conformation


causes-i) PH ii) temp iii) salt conc iv) detergant& other chemical which disrupt chemical bonds
